چکیده

The development of treatments to restore damaged cartilage that can provide functional recovery with minimal risk revision surgery remains an unmet clinical need. Gene therapy shows increased promise as innovative solution for enhanced tissue repair. Combination biomaterial approaches led the gene-activated scaffolds capable a sustained delivery therapeutic genes in strictly controlled manner be applied treatment number disorders. Within this study we developed novel microRNA (miR)-activated scaffold mesenchymal stem/stromal cells (MSC) chondrogenesis and repair through inhibitor miR-221 which is known have negative effect chondrogenesis. To fabricate miR-activated scaffolds, composite type II collagen-containing designed specifically were firstly manufactured by lyophilization then functionalised Glycosaminoglycan-binding transduction (GET) system nanoparticles (NPs) encapsulating inhibitor. Subsequently, cultured human-derived MSCs vitro under chondrogenic conditions 28 days. successfully transfected human cargo up silencing using promoted improved more robust cell-mediated response repressed early-stage events related MSC hypertrophy. Taken together, demonstrated capability enhance This thus demonstrates incorporation into might promising approach damage disease. article protected copyright. All rights reserved

Development of an antibacterial porous scaffold for bone defect treatment

Background & Aim: The use of bone scaffolds is one of the new and efficient techniques for repairing bone defects that provide a suitable platform for cell proliferation and growth to repair the target tissue. One of the most important causes of failure of transplants and surgical procedures is the invasion of bacteria at the site of the complication and the development of severe infection. The...

the aesthetic dimension of howard barkers art: a frankfurtian approach to scenes from an execution and no end of blame

Bioactive IGF-1 release from collagen–GAG scaffold to enhance cartilage repair in vitro

Tissue engineering is a promising technique for cartilage repair. Toward this goal, a porous collagen-glycosaminoglycan (CG) scaffold was loaded with different concentrations of insulin-like growth factor-1 (IGF-1) and evaluated as a growth factor delivery device. The biological response was assessed by monitoring the amount of type II collagen and proteoglycan synthesised by the chondrocytes s...

Matrigel scaffold combined with Ad-hBMP7-transfected chondrocytes improves the repair of rabbit cartilage defect

The aim of this study was to explore an effective method for the repair of cartilage defects using chitosan/glycerophosphate (C/GP) gel- and Matrigel-engineered human bone morphogenetic protein 7 (hBMP7)-expressing chondrocytes. Rabbit chondrocytes were obtained, cultured in vitro and transfected with an adenovirus containing hBMP7 and green fluorescent protein (Ad-hBMP7-GFP). The expression of...
